Properties of an Acidic Histone-binding Protein Fraction from Cell Nuclei
نویسندگان
چکیده
Calf thymus nuclei contain a histone-binding protein which specifically deacetylates histone Fractions F2Al and F3 by a hydrolytic cleavage of E-N-acetyllysyl residues. The protein has been purified over 500-fold by a combination of exclusion chromatography and isoelectric focusing techniques. It is acidic, as judged by its amino acid composition, and by its isoelectric point (pH 4.5). The protein contains tryptophan and is phosphorylated. Its molecular weight is estimated at 150,000 to 160,000. Histone-binding studies show that the isolated acidic protein selectively precipitates Histones F2Al and F3, both of which are substrates for the deacetylase activity. The isolated phosphoprotein attaches to chromatin fractions in vitro.

متن کاملPresence of multiple protein kinase activities in rat liver nuclei.
The phosphorylation of nuclear proteins has been suggested to play a role in the control of gene readout in mammalian tissues (1,2,3). Phosphorylation of histones has been shown to occur in rat liver in response to hormonal agents (4), and a histone phosphokinase has been isolated from calf liver (5). I.n addition, many of the non-histone (acidic) proteins of the nucleus are phosphoproteins. Th...

Background: Endometriosis (which affects up to 50% of infertile women) is one of the major causes impacting female infertility. Endometriosis, defined as the presence of endometrial glands and stroma outside the uterine tissue, causes a wide range of functional disorders in the process of follicular development and changes in the follicular milieu, resulting in the formation of an incompetent o...
متن کاملPredicting Protein Binding of Drugs Using Abraham Parameters: Effect of Ionization
Background and purpose: Protein binding (PB) is an important pharmacokinetic parameter in drug discovery and development. In past years Abraham parameters were used to predict some physicochemical and pharmacokinetic properties of drugs. But in these cases, the ionization of drugs in blood pH (7.4) was ignored. Recently, Abraham parameters of chemical compounds in ionized form are proposed. Als...
متن کاملRapid purification of HU protein from Halobacillus karajensis
The histone-like protein HU is the most-abundant DNA-binding protein in bacteria. The HU protein non-specifically binds and bends DNA as a hetero- or homodimer, and can participate in DNA supercoiling and DNA condensation. It also takes part in DNA functions such as replication, recombination, and repair. HU does not recognize any specific sequences but shows a certain degree of specificity to ...
